擅長核酸編輯和組裝,致力于開拓DNA在納米材料、生物傳感、臨床診斷等方面的基礎(chǔ)和應(yīng)用研究,開展的方向包括:
對靶點或標(biāo)志物明確的重大惡性腫瘤和慢性疾病,開發(fā)可用于體液診斷的便捷適配體探針(試劑盒);對于標(biāo)志物未知的重大惡性腫瘤和慢性疾?。ㄖ饕侨幮匀橄侔?,以適配體探針庫為抓手,通過臨床樣本間的對沖抵消,篩選鑒定疾病相關(guān)的標(biāo)志物和對應(yīng)的檢測探針,揭示疾病的分子機(jī)制并明確藥物可作用的靶點;
開發(fā)新型3D框架核酸的設(shè)計和組裝方案,在保持組裝體近原子分辨率的前提下,將自組裝的尺度在3D空間從納米推向微米和亞毫米級,以滿足其作為介質(zhì)在材料科學(xué)中的更多應(yīng)用需求;同時開發(fā)活細(xì)胞生產(chǎn)制備框架核酸的生物技術(shù),降低制備成本,為框架核酸的臨床研究提供支持;
利用試管內(nèi)進(jìn)化的高效篩選策略分離獲得重大疾病相關(guān)的核酸酶(DNAzyme和Ribozyme),將其與框架核酸融合,編輯組裝“環(huán)境響應(yīng)型智能納米藥物載體”,用于重大疾病的靶向精確遞藥。
